The conditions of cell cultivation have to be summarized in order to provide consistency independent of the day and the individual cell culturist. The culture conditions should be adapted for each individual cell type. Cell banks are a reliable source of information about the appropriate growth medium for a given cell line. The medium composition should be listed in detail including all ingredients such as basic medium, serum and supplements, or additives necessary for growth or differentiation of cells. Certain cell lines may require a certain growth surface / matrix, which should be consistently used during cultivation. Recording the dissociation method finally enables comparable sub-cultivation independent of the person working with the cells.
Evaluation of the doubling time defines the optimal time point for passaging as well as cryopreservation of cells. Cells should be cryopreserved when they are actively growing in their log phase. Documentation of freezing medium, the average freezing number as well as the appropriate thawing density of cells further facilitates a comparable routine handling in cell culture.
